Thesis Title : Determination of the Sun Protection Factor (SPF) of Black Turmeric (Curcuma caesia Roxb.) Rhizome Extract Based on Solvent Variation Using the UV-Visible Spectrophotometric Method

Indonesia, as a tropical country, is exposed to high levels of ultraviolet (UV) radiation, which increases the risk of various skin disorders. The Sun Protection Factor (SPF) is used to measure the effectiveness of sunscreen in protecting the skin from UV radiation. Black turmeric (Curcuma caesia Roxb.) has potential as a natural sunscreen due to its content of flavonoids, phenolic compounds, and curcumin, which exhibit photoprotective properties. This study aimed to determine and compare the SPF values of black turmeric rhizome extracts prepared using different solvents, namely methanol, 70% ethanol, and 95% ethanol, employing the UV–Vis spectrophotometric method. This research was conducted using an experimental design. The study procedures included plant identification, preparation of simplicia, determination of drying loss, extraction by maceration, determination of moisture content, phytochemical screening, and in vitro SPF evaluation using a UV–Vis spectrophotometer at wavelengths of 290–320 nm. The results showed that the methanolic extract exhibited SPF values ranging from 34.89 to 37.32, classified as high protection, while the 95% ethanolic extract showed SPF values ranging from 31.25 to 37.10, also categorized as high protection. In contrast, the 70% ethanolic extract exhibited SPF values ranging from 20.46 to 27.49, which were classified as moderate protection. Statistical analysis indicated that the data were normally distributed and homogeneous, allowing further analysis using the One-Way ANOVA test. The One-Way ANOVA results showed a p-value of 0.003 (< 0.05), indicating a statistically significant difference in the SPF values of black turmeric rhizome extracts based on the extraction solvent used. The Post Hoc Tukey test revealed that the 70% ethanolic extract differed significantly from both the 95% ethanolic and methanolic extracts, whereas no significant difference was observed between the 95% ethanolic and methanolic extracts in terms of SPF values.
